I have been to the graveyard district at night and can't find Valens Mistress anywhere although I havm't been underground yet. I am in Chapter 3 and I suspect she is a Vampire so does it matter that I have already killed some vampires in that attack at night time?

Have you sided with someone yet? If you have that could explain why you can't find her there. She should be right at the entrance.
