The "use device" mouse pointer in Baldur's Gate 2 irritatingly enough shares almost the same icon as the "can't walk here" mouse pointer. Make sure you don't let your eyes deceive you.

If you mouse over a coffin that contains a vampire, the mouse icon will turn into a sword. If you click the coffin it will give you a message saying "... the vampire is now truly dead". If the mouse icon does not turn into a sword, then you haven't killed all the vampires (i.e. the one that is supposed to return to the coffin). Remember that you have to be standing next to the coffin to make it work, and once you've staked a coffin it won't let you stake it again.

Just let the story unfold before you. Nothing beats playing the game yourself without having been spoiled about what's happening. Your first time through any game you find things out the hard way. It only means you can find out new things the second time.

She's kind of difficult to see... Go to the left of where you first came to the city... but she's worth it, as she has absolutely no economic sense. Maybe the economy is different there and the gold piece is stronger than the, um, shark tooth or drow skull or whatever it is they use for currency.
