This could be a movie and/or a series. My vote would have to go to Goldfinger. My favourite series would be South Park, its just too funny

The Carol Burnett Show.
Twenty years later, I can still laugh my a$$ off.

Shows like "Whose Line Is It Anyway" could really take some lessons from Carol Burnett on what true and hilarious improv is all about.
I don't believe for a second that anything in "Whose Line" is not scripted.

It's quite obviously "Only Fools and Horses". It's shown all around the world and is still hilarious to this day.

X-Files, Stargate SG-1, JAG, Hogan's Heroes but I have to say my absolute favourite TV show is Buffy the Vampire Slayer. You can all stop laughing mockingly at me now. My favourite movie is Gone in Sixty Seconds.
