Yes you must go through the Shrine of Anephas first. Also, there is a bug in the game, the quests are all out of order for the Brimloch Roon Wizards Guild, check this forum.

There are actually three ways to get to the Lost Sea.

First, go through Anephas' Shrine. That's the *right* way.

Second, you can vanish through the trees in the clearing in front of Anephas' Shrine. There's a spot on the northern edge, along the western side, IIRC. Radek once posted explicit details... you can search for a post from him.

Third, you can tree-surf to get over there. Search the forum for Tree-surf and you'll find details. It can be frustrating, but it works.

And Shadow Stranger is right about the Wizards Guild quests. Opening the Shrine is the third quest, and the objects you need to satisfy the second quest are most often on the other side. You can sometimes get them on the Enchanted Sea side, but you end up relying on random encounters. Or attacking Oakenmir and hoping he brings reinforcements...

See Page 9 of the Best Tips post for the description of the Wizards' quest problem. One solution is to hex-edit the seeds into the inventory of a PC, see the post "Wyvern's Emporium of Lost and Unobtainable Items!". You want the Treant Seed item.
