As with all characters, when you click on them, you get the little menu at the bottom to use their spells, weapons, whatever. Well, when I click on Minsc or Mazzy, there's a button shaped like a shield and when you mouse over it, it says Guard. What does this do?

I think it is/was intended to put your characters in a more alert state and to alter their AI scripts but I never use it and there is not even a proper reference in the manual (it only says that it is there on some occasions but does not explain it in the button listings like it does with thieving or turn undead)
IIRC there is an explanation in the tutorial but I can't check that out because I have BGT installed.

It makes a character protect an ally. Only realy useful if the one being guarded is a major tank, and the one doing the guarding uses a ranged weapon... and then it depends on terain...

This may also help, from the BGII Quick Reference Card: Guard/Defend: Left click on button, left click, hold and drag circle around area for character to defend/guard. Could come in handy, I guess, although I'm sure LennonCook's comments are still valid.
