Can the samurai be your main unlocker? If so when or how can he/she learn the lock/trap skill?

Samurai do not have an innate lock/trap skill. They can only aquire the skill by either changing classes or using items such as the poseur's cap or thieve's buckler, which would give them a total of l/t skill of 20. Only rogues, ninjas, bards, and gadgeteers have an innate lock and traps skill, which they can begin learning at character creation.

He has got no lock/trap skill! But he could use this green cap you find at the lower monastery, that might be enough combined with someone casting knocknock? But personally i wouldn't recommend that. Only rogues, ninjas, bards and gadgeteers have that skill.

I have played pure spellcaster parties, so i know it is possible without the lock/trap skill. But you should be able to cast knock-knoch with 3 characters (in ironman mode - with reloading, this might differ). If only one character has the spell, keep (and buy) any knock picks you find.

Samurai's can't learn Locks & Traps but they can learnt he Knock-Knock spell. I've found that the cap you find in the monastery combined with one or two Knock-Knock spells will get you through the game no problem, though Ironman may be a bit tricky.
