1. In fact, there is no cleric's stronghold in the game - only a set of quests.
2. Your protagonist should be a single class cleric. A multiclass or a dual class clerics need not be eligible for the cleric's stronghold. I am not sure what rules are used in deciding which stronghold will be given to a multiclass or a dual class character but I am sure that, for example, the dual class ranger/CLERIC gets the ranger's stronghold.
Now, it depends on your alignment. If you are LG, NG or CG then check the Temple of Lathander. If you are LN, TN or CN then check the Temple of Helm. If you are LE, NE or CE then check the Temple of Talos. The quests will differ a bit according to your alignment. If you want to be successful with the quests then always decide according to your alignment during the quests. Even if the decision does not fit your feeling.

If you're multiclassed, you can choose a stronghold. The first one you get to is yours.

One other detail abot getting the Cleric Stronghold... You have to go to the Temple District, then head towards the middle of it. After you hear the dialogue about "join the Cult of the Unseeing Eye!" you will automatically be approached by a member of your faith (based on your alignment). You can complete the quest you get even if you aren't a Cleric, but only after completing it will you be invited to "join the faith", & thus gain the Cleric's Stronghold.
