It depends on how you are setting up to run DOS. Some programs have to be run in DOS only. Assuming you don't have Windows ME, you could start this by shutting down into DOS mode. You will need to setup the EMS with a progam like EMM386 or an equivalent. If you can run the game in a window that is running dos, you need to configure the properties of the icon that takes you to DOS to enable the memory specifications that you want. I don't have DOS on my computer so can't walk you through it exactly, but when you select properties, you should see many options listed that are kinda tiled at the top. You just go through them til you find the one that relates to memory. Go ahead and set it up for Expanded memory and then exit and save. When you double click on the item to start the game, it will then use those properties. I'll see if I can find a better answer for you, but that's basically what you need to do. If you create a startup disk from Windows, it will normally give you most of the options that you will need. This may work better as it will take you out of WINDOWS altogether.
