The Twisted room house isn't a quest. It might have been though to be one but never put in, but there isn't really a quest to it. You simply have to have a rogue stone in order to ender the Twisted runehouse. If you try to enter the door without having a roguestone, it just won't let you in.
There are some Twisted rune guys outside of the house you run into during the game so maybe they were part of a broken quest somewhere.
The Twisted room house is in the bridge district. Its a pretty hard battle, so make sure you save before you go in. The best mage staff in the game is in there, so if you have a mage, you might want to get it.
