Yes, in order to have Yoshimo's heart (by bringing it to Temple of Ilmater will grant you 200k exp points), you will need to kill him after you beat Jon in his lab in Spellhold and YES, YOU WILL NEED TO TAKE HIM INTO SPELLHOLD WITH YOU. If it is exp point you want, you will get plenty later on, probably not worth the trouble reloading.
