Hmmm... interesting.

The only way I can think of to become an assassin twice in the same game is to be playing unpatched, get the dagger, and hand it off to someone else. Then to go back in with it... I haven't played unpatched in a long time, so I'm not sure whether the checks in the game act based on whether you answered "No" to the promotion chance, or whether you answered at all.

In any case, I believe you'd just start over as a level one assassin. No big deal, just like any other class change.
