While you will meet up with Kol later in the game, if he's left the Serpent Temple area before you got the key, you cannot get the key UNLESS

you have a save game from shortly before the time you meet him,

revert back to the last time you were in town IF that was before you met Kol,

OR you can hack yourself a key (I can give you instructions on how to do that)

But it really isn't that important that you have the key. I've played a couple of games where I never bothered with it at all.
