If you are referring to the door with a "level 12 lock" ie the stained glass door in the lich's room, then you can:

1)accept Adrianna's quest and ask for the treasure as the reward
2)have really good lockpicking skills (e.g. if your pick lock skill is only 7, then buy a level 5 lock pick set, or get your lock pick skill to the max of 12)..

Inside the room there is one treasure chest with a few goodies, which are nice to have but not essential to completing the game..
