I don't think Shemeesh or the villagers will disapear if your rest after killing the Lizard King with your Cleric/Ranger. You'll fully recover in 16 hours, which is a short enough time. If you're worried, you can leave and rest outside the DE entrance. Then come back.
I once returned to DE after finishing Severed Hand to pick up some items I purposely left in a chest, and the villagers were still there!
