First of all, it is the Twisted Rune, not the twisted ruins.

This isn't really much of a quest, it is just a powerful multi-race party you encounter. To meet them, you must first of all have a Rogue stone in your inventory. Now, go to the Bridge District, and to the house where the people who buried Tirdir were located (down south-west). Next to it, there is another house with two doors. Behind one is a Lich, behind another is the Twisted Rune. Pick or bash the lock on the door which is turned against the ocean. With the Rogue stone in the inventory, you can now enter there.
