but I did not find a Vampire
Did I bypass her??/
It is daylight
do I need to wait till night???

Lysandra is still there. Pickpocket her if you don't want to become a vampire.

When you first come up from the catacombs sewer, she's on the first landing, near a coffin and several skulls. She's friendly...a little TOO friendly, actually. Watch out!

Day or night - doesn't matter - she's there.

Since you managed to get all the way up to D'Soto's tower to get a relic, you must have passed her landing along the way and just not noticed her. Halfway up the staircase!
